血小板蛋白S (PSplt)调控大静脉血栓中的血小板活化和凝血,但不调控大动脉血栓中的凝血。 PSplt可限制血栓中的凝血酶生成,并确保高度活化的血小板和纤维蛋白局限于损伤部位。

血小板(PSplt)中的抗凝蛋白S (PS)类似于血浆PS,在血小板激活时释放;但其在血栓形成中的作用尚未得以阐明。

在本研究中,研究人员通过Pf4-Cre转基因(pro1lox/loxPf4-Cre+)使小鼠PSplt表达失活,促进剪变率较低的腔静脉产生血栓倾向,但不影响剪变率较高的颈动脉。

剪变率较低时,PSplt发挥活化蛋白C和组织因子通路抑制剂的辅助因子的作用,限制生长血栓中X因子的活化和凝血酶的生成,并确保高度活化的血小板和纤维蛋白局限在损伤部位。在凝血酶浓度较高的情况下,来自pro1lox/loxPf4-Cre-小鼠的血凝块收缩,而来自pro1lox/loxPf4-Cre+小鼠的血凝块不会收缩,因为纤维蛋白网络高度密集。

因此,PSplt可以调控血小板的活化以及大静脉血栓中的凝血,而在大动脉血栓中则无此功能。
