Science:科学家发现克服遗传不育症的新技术

正常数量的染色体对于个体的正常发育和健康都至关重要。性染色体异常是最常见的人类不育症遗传因素。其中,性染色体三倍体大约影响人群的0.1%。在XXY和XYY综合征中,精子发生分别会被过量的X和Y染色体所阻碍。XYY的男性经常发生多余性染色体的自发丧失,因此通常是可育的。但是在XXY的男性中,多余性染色体的自发丧失就比较少见。

正常数量的染色体对于个体的正常发育和健康都至关重要。性染色体异常是最常见的人类不育症遗传因素。其中,性染色体三倍体大约影响人群的0.1%。在XXY和XYY综合征中,精子发生分别会被过量的X和Y染色体所阻碍。XYY的男性经常发生多余性染色体的自发丧失,因此通常是可育的。但是在XXY的男性中,多余性染色体的自发丧失就比较少见。 来自弗朗西斯克里克研究机构的科学家们发现,在重编程成为诱导多功能干细胞(iPSC)的过程中,来自无菌XXY或是XYY小鼠的成纤维细胞会发生三体偏倚染色体丢失的现象,从而丢失额外的染色体。通过该现象能够产生整倍体XY的iPSC,从而可以分化成为男性生殖细胞谱系,并进一步成为具有功能的精子。通过细胞质内精子注射的手段,XXY和XYY的小鼠就能够产生染色体正常,可育的后代了。在小鼠XX和XY iPSC生成期间,性染色体丢失相对不频繁。 三体偏倚染色体丢失也适用于其他染色体,比如从唐氏综合征小鼠模型的细胞中产生整倍体iPSC。它还可以从人类三体患者成纤维细胞中产生整倍体iPSC。 这些发现有利于不育症以及其他三体表型相关疾病的治疗。 原始出处: Takayuki Hirot
