代谢组学分析可能通过测量伴随癌症进展的代谢物变化来确定治疗耐药的新治疗靶点。研究人员之前对肾细胞癌(RCC)进行了整体代谢组学(G-Met)研究,发现了可能参与RCC中舒尼替尼耐药的代谢物。

最近,研究人员通过细胞内代谢物来阐明RCC中舒尼替尼耐药的可能机制。研究人员从RCC细胞(786-O)注射小鼠的肿瘤组织中建立了舒尼替尼耐药和对照RCC细胞系,量化了G-Met研究中发现的特征代谢物,并利用液相色谱-质谱法比较两种细胞系之间的细胞内代谢情况。结果表明,舒尼替尼耐药的RCC细胞系与对照组RCC细胞系相比,在舒尼替尼暴露下,耐药RCC细胞系表现出显著的蛋白激酶B(Akt)去抑制和间质到上皮转化(MET)磷酸化。在鉴定的代谢产物中,谷氨酰胺、谷氨酸和α-KG(参与谷氨酰胺摄取进入三羧酸(TCA)循环进行能量代谢);果糖6-磷酸、D-琥珀糖7-磷酸和葡萄糖1-磷酸(参与增加糖酵解及其中间代谢产物);谷胱甘肽和肌醇(抗氧化作用)在耐舒尼替尼的RCC细胞系中显著增加。特别的是,谷氨酰胺转运体(SLC1A5)在舒尼替尼耐药RCC细胞中的表达与对照相比显著增加。

最后,研究人员指出,他们揭示了能量代谢与谷氨酰胺摄取和糖酵解上调,以及抗氧化活性与RCC细胞的舒尼替尼耐药性有关
