在本研究中，研究人员使用人骨髓基质细胞和MC3T3-E1前成骨细胞的细胞培养系统，结果显示：1）褪黑激素诱导正常和炎症条件下的骨形成; 2）Wnt4对炎症刺激中褪黑素诱导的骨形成至关重要; 3）褪黑激素和Wnt4诱导的骨形成是通过与不同的卷曲LDL受体相关蛋白复合物相互作用激活β-连环蛋白和p38-JNK MAPK途径而发生的; 4）褪黑素以Wnt4依赖的方式抑制NF-κB对成骨的抑制作用; 5）褪黑激素通过ERK1/2-Pax2-Egr1途径诱导Wnt4表达。

总之，我们展示了褪黑激素诱导的炎症环境中骨形成的新机制。褪黑激素诱导的Wnt4表达对于其骨诱导作用和NF-κB对骨形成的抑制作用是必需的。我们的新发现可能为其潜在的转化应用提供有用的信息。
