两家基因编辑公司强烈反击批评CRISPR导致突变的文章，称应将其撤稿

上周，《自然方法》杂志发表了一篇对CRISPR技术的批评文章，声称基因编辑工具CRISPR已经在小鼠基因组中引发了“意想不到的突变”（详细见：Nat method：CRISPR基因编辑技术导致小鼠体内上百种非目标基因突变）。该文章的发表，导致两家基因编辑公司股票的暴跌：Intellia Therapeutics和Editas Medicine。近日，两家公司对该文章表示强烈不满，认为其结论完全错误，甚至就不应该公布。

Intellia公司的首席执行官呼吁将该文章撤稿，并将其从科学文献中删除。

他说：“这份出版物已经受到了媒体和公众的巨大关注，对我们造成了重大损失。鉴于该文章实验设计和结果讨论的问题，我认为《自然方法》杂志应该将其撤稿。”

《自然方法》出版方发言人则表示，他们已经收到了一些关于该文章的反馈，他们正在认真考虑，并与作者讨论相关问题。该文的作者并没有直接做出回应。

《体内CRISPR-Cas9编辑引发的不可预测基因突变》一文声称基因编辑工具在编辑小鼠基因过程中造成了广泛和不可预知的破坏，引入了数百种意外的基因突变。文章一经发表马上成为头条新闻，CRISPR概念股随后股价大跌。其中，Editas医药，Intellia Therapeutics和CRISPR治疗公司的股票市场总价值高达10亿多美元。


在Twitter等社交平台上，一些科学家很快指出了该文的部分基本错误，包括错误的基因识别、数量偏少的动物实验，最严重的是，该文将动物的正常遗传差异当作是CRISPR编辑带来的结果。Editas首席技术官与其它11位科学家联名写到，该文实验数据不能支持其结论。这些科学家中还包括来自哈佛大学教授。他们认为该文章“很有可能”被撤销，因为文章忽略了很多关键问题。

CRISPR技术被誉为“革命性的基因工程手段”，但有时它的作用会被夸大，包括声称这个技术可以会治愈所有遗传疾病，并解决世界的食物短缺问题。事实上，CRISPR的确可以剪切特定的DNA序列，从而修正或改变基因。虽然其功能强大，但如果基因组中其他位置有相同或相似的DNA序列，则可能导致更多DNA的随机改变。很多人都表示过对该技术潜在副作用的担心，其中甚至包括该技术的发明者们。

随意将CRISPR用于治疗可能是非常危险的。如果一个目标基因片段也同时处于重要的基因中，一个错误的基因切除可能是灾难性的，但许多科学家认为，找准基因片段可以消除大部分风险。

CRISPR的易用性意味着几乎任何实验室都有条件使用。中国已经开始在人类上进行实验，但这种操之过急的研究可能回让出错的可能性增大。 因此，Editas推迟了关于CRISPR用于治疗眼部疾病的研究。

然而，Intellia认为，该文章作者无视了CRISPR的很多研究成果，这些作者即不是CRISPR 的专家，也不是全基因组测序或者基因遗传学家，因此这种“意想不到的突变”的结论缺乏科学性。
