最近,研究人员对435,563名欧洲血统的个体进行了PAU的代理表型荟萃分析,结合酒精使用障碍和问题饮酒。

研究人员发现了29个独立的风险变异,其中19个是新的。PAU与138个表型有遗传相关性,包括物质使用和精神特征。

在一个独立的生物库样本(BioVU,n = 67,589)中进行的全表型多基因风险评分分析证实,PAU与药物使用和精神疾病之间的遗传相关性。PAU的基因遗传性在大脑以及保守和调节基因组区域富集。

孟德尔随机化表明,物质使用、精神状态、冒险行为和认知表现对PAU有因果效应。

总之,这项大型的PAU元分析确定了新的风险位点,并揭示了与许多其他特征的遗传关系。
