目前已知胆固醇是多种类型离子通道的重要调节因子,但其与离子通道相互作用的具体机制和动态变化信息仍所知甚少。既往曾有研究显示,Kir2通道对于胆固醇的敏感性是通过与通道蛋白上的“胆固醇敏感”区域的直接相互作用而实现。

在当前这项研究中,来自美国伊利诺伊大学芝加哥分校的Nicolas Barbera博士及其同事使用Martini粗粒化动力学模拟方法,通过将Kir2.2通道植入一个包含POPC磷脂和30mol%胆固醇的膜模拟体系,分析了胆固醇与Kir2.2通道在长时间跨度中的动态变化。这一方法可以动态、无偏倚的模拟胆固醇分子从脂质膜环境移行至Kir2.2蛋白质表面的情况,从而可以探索胆固醇在通道表面位置及内部发生相互作用的有利度。

研究者发现,Kir通道周围的胆固醇环境会形成一个发生不同类型短期和长期相互作用的复杂环境,多种胆固醇分子同时与离子通道发生相互作用。根据网络理论原理,研究者在既往已经发现的胆固醇敏感区域中,确定了存在4个依赖于离子通道打开或关闭构象状态的不连续胆固醇结合位点。

此外,研究者还发现,脂质膜中的胆固醇水平降低2倍(既往显示可以增加Kir2活性),导致这些离散位点在离子通道打开或关闭状态时均出现位点特异性胆固醇占有率降低:最深位点的胆固醇分子占有率无变化,而其他位点均呈大幅增加趋势。

该项研究的资深作者,来自美国北卡罗来纳州立大学分子生物医学系的Akpa BS教授总结支持:“胆固醇本身并不是一个有害的事物,它始终存在于细胞膜内。但当胆固醇水平升高时,就会导致出现问题。鉴于正常细胞膜中的胆固醇比例约为30%,我们希望了解为什么当这一比例仅小幅升高时(从30%升高至40%),就会使情况突然变得严重。当前的研究的结果表明,胆固醇或许实际上应该在上述4个位点中被‘锁定’,但我们发现它们会试图移行至可能尚未被‘占领’的地方,而这会干扰蛋白质通过改变其形状来打开或关闭离子通道的能力。由于细胞需要通过离子通道来调节不同时间中的离子进出这一复杂变化,问题也就随之出现。尤其重要的是,这可能会导致正常的离子交换顺序发生混乱,插入错误信息。”
