红细胞含有独特的膜脂群。虽然我们对蛋白表达调控知之甚多,但对红细胞生成过程中的脂质代谢调控的研究缺极其匮乏。近日,Blood杂志上发表一篇相关文章,研究人员发现在,在人类和小鼠的红细胞生成的终末阶段,介导胆碱磷酸水解成胆碱的磷酸乙醇胺和胆碱磷酸酶——PHOSPHO1的转录水平明显上调,伴随卵磷脂和胆碱磷酸分解代谢增加,与人和小鼠终末红细胞生成的整体脂质分析一致。

敲除PHOSPHO1可破坏胎鼠和人类有核红细胞的分化,而敲除成年鼠的PHOSPHO1会损伤苯肼诱导的应激性红细胞生成。丢失PHOSPHO1也可破坏小鼠胎肝祖细胞的胆碱磷酸代谢,导致多种脂质累积;氧化磷酸化水平降低,导致ATP产量减少。在PHOSPHO1敲除的有核红细胞中,糖酵解(无氧)代替了有氧磷酸化,而且糖酵解水平增加用于合成丝氨酸和甘氨酸。

本研究阐明了脂质代谢在红细胞终末生成过程中的动态变化,并揭示了磷脂酰胆碱和胆碱磷酸代谢在能量平衡和氨基酸供应中发挥关键作用。
