J Exp Med:烟酰胺单核苷酸转移酶(NNT)通过维持Fe-S蛋白功能而调节NSCLC线粒体代谢

人类肺癌表现出强大而复杂的线粒体代谢,可能是由肺组织的高氧性造成的。由于活性氧(ROS)的生成是这种代谢的副产物,因此需要以烟酰胺腺嘌呤二核苷酸磷酸(NADPH)的形式来降低氧化应激,以应对这种线粒体

人类肺癌表现出强大而复杂的线粒体代谢,可能是由肺组织的高氧性造成的。由于活性氧(ROS)的生成是这种代谢的副产物,因此需要以烟酰胺腺嘌呤二核苷酸磷酸(NADPH)的形式来降低氧化应激,以应对这种线粒体活性的增强。烟酰胺单核苷酸转移酶(NNT)可通过NADPH的生成来维持线粒体的抗氧化能力,但其在非小细胞肺癌(NSCLC)中的作用尚未明确。本研究发现NNT的表达在肺癌小鼠模型中明显增强了肿瘤的形成和侵袭性。另外,进一步研究发现NNT缺失会导致线粒体功能障碍,这与氧化应激的大量增加无关,而是以铁硫蛋白的活性降低为标志。这些缺陷与NADPH的有效性和ROS的积累有关,这表明NNT在减轻这些关键蛋白辅因子的氧化中起着关键作用。

实验设计

许多常见的肺癌小鼠模型是利用C57BL6/J小鼠的饲养策略而建立。有趣的是,这些小鼠携带纯合外显子7-11的框内缺失以及线粒体中Nnt基因序列的错义突变导致非功能蛋白的表达。因此,可以利用这种自然缺失的等位基因(NntΔex7-11)来评价NNT对肺癌发生的影响。

首先,使用Kras突变(LSL-KrasG12D/+)引起的肺癌模型来检测Nnt表达对肺癌发生的影响。携带Cre重组酶的腺病毒感染LSL-KrasG12D/+小鼠,可诱导KrasG12D在肺上皮中的表达和肺腺癌的形成。利用LSL-KrasG12D/+小鼠和C57BL6/J小鼠交配产生Nnt+/+表达小鼠或NntΔex7-11/Δex7-11表达小鼠(图1A-1B)。该模型中Nnt的表达导致Cre重组酶诱发肿瘤的负荷加快3个月(图1C-1D)。然后,利用Kras突变与p53缺失同时表达(LSL-KrasG12D/+;Trp53flox/flox,也称为KP)建立肺癌模型。Nnt的表达没有改变Cre诱导的KP小鼠的存活率(图1E)。有趣的是,在这个模型中,p53的缺失消除了Nnt表达对肺癌形成的影响,并且无论Nnt表达如何,实验最终都存在明显的肿瘤负荷(图1F)。肺癌负荷比例的量化在不同基因型小鼠之间没有差异(图1G-1H)。

虽然不同基因型小鼠之间的肿瘤负荷没有差异,但确实观察到模型中肿瘤侵袭性的差异。我们发现Nnt+/+小鼠中51.3%的肿瘤为3级(腺癌)或以上,而NntΔex7-11 / +和NntΔex7-11 /Δex7-11小鼠中仅有36.5%和38.8%的肿瘤是高级(图1I)。Nnt+/+小鼠发生4级肿瘤的频率显著增加,证明了肿瘤侵袭性的变化(图1J)。总之,这些数据表明Nnt促进了肺癌的发生和侵袭。

2 NNT的缺失不影响线粒体硫氧还蛋白(TXN)抗氧化体系

为了进一步评估NNT对肺癌生物学的影响,首先评估shRNA干扰NNT蛋白的表达,包括4个表达NNT蛋白的NSCLC细胞系 (A549, H1299,H2009和PC9)和不表达NNT蛋白的H441细胞(图2A)。干扰NNT蛋白表达抑制NSCLC细胞的增殖能力(图2B)。并且发现慢病毒感染4天后NNT的下调降低了H2009和PC9细胞的生存能力(图2C)。此外,H441细胞的增殖不受NNT的影响。

通常认为NNT有助于NADPH的还原能力,从而维持线粒体蛋白抗氧化系统处于还原状态(图2D)。干扰NNT降低了H1299、H2009和PC9细胞中NADPH: NADP+的比例 (图2E)。慢病毒感染4天后线粒体中H2O2显著增加(图2F),所以NNT对H2O2解毒过程很重要。

为了确定这些线粒体ROS的增加是否与线粒体抗氧化系统有关,于是利用Western blotting评估线粒体中过氧化物蛋白3(PRDX3)的氧化状态。H2O2通过PRDX3解毒,即一对半胱氨酸二硫键诱导PRDX3形成二聚体,并且必须通过TXN还原这些半胱氨酸二硫键才能还原PRDX3的抗氧化功能。PRDX3二聚体的积累是PRDX3氧化的标志,是线粒体氧化应激的常用标志物。

奥拉诺芬是一种TXN还原酶(TRXR)抑制剂,虽然奥拉诺芬的治疗会导致PRDX3蛋白大量氧化,但NNT的缺失并没有增加PRDX3的氧化(图2G)。此外,NNT的缺失也没有降低线粒体TXN2或TRXR2的蛋白水平。总之,这些结果表明NNT在NSCLC细胞增殖能力上发挥重要作用,但NNT活性的丧失并不会损害线粒体TXN抗氧化系统,也不会引起明显的氧化应激。

3 NNT的缺失会损害线粒体的氧化能力

鉴于NNT在IMM中的定位及其影响膜上质子通量和还原能力,于是试图探讨NNT对线粒体氧化代谢是否重要。首先,使用海马细胞外通量分析仪进行线粒体压力测试,评估NNT缺失对一般线粒体氧化功能的影响。结果发现NNT缺失细胞的耗氧量(OCR) 随线粒体抑制剂的依次传递而被减少(图3A)。值得注意的是,NNT缺失细胞的最大呼吸能力在不依赖于非耦合呼吸的情况下显著降低(图3B)。这提示线粒体氧化缺陷与NNT对质子梯度的影响无关。鉴于NNT缺失明显影响呼吸能力,于是检测NNT缺失的H441细胞是否比NNT表达细胞具有更少的氧化和更强的糖酵解能力。这说明这些细胞在没有NNT的情况下可能通过线粒体NADPH来源而维持线粒体的功能。

线粒体的氧化代谢依赖于ETC的功能,ETC由介导电子传递的Fe-S蛋白复合物组成。考虑到Fe-S蛋白复合物的生物合成过程需要NADPH,于是研究NNT干扰后线粒体呼吸功能的下降是否与Fe-S蛋白相关。线粒体压力测试可以对呼吸功能进行一般性的分析,但不可以对单个ETC复合物成分进行评估。因此,作者进行了更为专业的实验来分析Fe-S蛋白依赖的呼吸相关复合物(I, II, III)。结果发现,基于复合物I(丙酮酸和苹果酸)的摄取,在NNT干扰后复合物I-III的活性显著降低(图3C)。此外,NNT缺陷细胞明显降低了琥珀酸的OCR,表明降低了琥珀酸脱氢酶(SDH)活性和复合物II-III流量(图3D)。导致这一现象的重要原因是SDH在ETC和TCA循环中扮演着双重角色。

除了通过ETC维持电子流量外,Fe-S蛋白还维持对氧化代谢至关重要的其他蛋白酶的功能。为了确定NNT是否参与其他Fe-S蛋白的功能,作者评估了TCA循环中Fe-S蛋白——顺乌头酸酶(ACO2)的活性。结果发现干扰NNT显著降低了NSCLC细胞系中ACO2的活性(图3E)。ACO2活性的降低可能会破坏TCA循环,进而减少驱动ETC流量的还原性物质的生成。为了补充非小细胞肺癌中ACO2功能的分析,于是评价Nnt表达对KP肺癌中ACO2活性的影响。结果表明,Nnt+/+小鼠肿瘤中Aco2活性明显高于NntΔex7-11/+ 和NntΔex7-11/Δex7-11中的,缺乏Nnt的肿瘤表现出最低的活性(图3F)。

4 外源性NADPH在NNT缺失后维持Fe-S蛋白功能

为了确定与干扰NNT相关的线粒体Fe-S蛋白功能的降低是否与NADPH:NADP+的降低有关,我们试图提供一种线粒体NADPH的外源性来源。为了实现这一目标,我们选择了酵母线粒体中NADH激酶pos5p,它可以磷酸化NADH而产生NADPH。虽然pos5p以前在细菌中也有外源表达,但据我们所知,pos5p还没有被引入哺乳动物系统。为了检测我们是否能够有效地在人类NSCLC细胞的线粒体中表达pos5p蛋白,我们对酵母蛋白进行了修饰,使其包含HA-tag。Western blot结果显示,在H1299、H2009和PC9细胞的线粒体中成功表达了HA-tag标记的pos5p蛋白(图4A)。选择这些细胞系是为了评估pos5p修复和NNT缺失相关的Fe-S蛋白缺陷的能力,发现干扰NNT后对它们造成很严重的影响。

pos5p的表达挽救了细胞中干扰NNT后引起的NADPH: NADP+比例的减少(图4B)。这与表达pos5p细胞中pos5p被干扰后呼吸链复杂活动的减少有关(图4C和4D)。此外,pos5p的表达完全挽救了NNT干扰后ACO2活性的下降(图4E)。总之,这些数据表明维持NNT表达缺失时NADPH的水平可以保护NSCLC细胞中Fe-S蛋白的功能。

5 NNT的缺失不会破坏Fe-S蛋白的生物合成

考虑到线粒体外源性NADPH能够减弱NNT缺失相关Fe-S蛋白的缺陷,并且NADPH是高效维持Fe-S蛋白生物合成的必备条件,因此我们接下来试图确定NNT活性是否维持这一过程。Fe-S蛋白生物合成发生在一个由半胱氨酸脱硫酶(NFS1)和Fe-S支架蛋白(ISCU)组成的多蛋白复合物中。复合物中任何一种成分的缺失都会影响生物合成,并且也与线粒体缺陷相关。因此,我们干扰NFS1或ISCU表达来研究破坏Fe-S蛋白生物合成后对呼吸链和ACO2的影响。

在H2009细胞中,干扰NFS1后显著降低丙酮酸/苹果酸反应和琥珀酸反应中呼吸链复合物I-III的活性,而在PC9细胞中只有复合物II-III活性被明显降低(图5A和5B)。另外,两个细胞系中,在丙酮酸/苹果酸和琥珀酸的作用下,干扰ISCU表达后显著减弱了OCR(图5A和5B)。并且干扰NFS1或ISCU后显著降低了细胞系中ACO2活性(图5C)。有趣的是,NNT干扰后所引起的缺陷与Fe-S蛋白生物合成所需物质造成的缺陷是同等重要的(图5A-5C)。

为了证明Fe-S蛋白缺陷对NSCLC细胞的线粒体代谢有功能性影响,我们对干扰NNT或ISCU细胞进行基于液相色谱-高分辨质谱(LC-HRMS)的代谢组学研究。 对这些细胞TCA循环的代谢物进行分析,结果显示大多数中间产物的丰度发生了显著变化(图5D)。这些都表明了氧化代谢的严重破坏和Fe-S蛋白功能缺陷的一致性。具体地说就是干扰NNT和ISCU表达后,丙酮酸、苹果酸和富马酸被消耗。NNT缺失细胞中的柠檬酸水平被耗尽,但干扰ISCU细胞中并没有此结果。另外,干扰ISCU细胞中琥珀酸大量积累,而在NNT缺失细胞中则不存在此现象(图5D)。

除了Fe-S蛋白ACO2和SDH外,TCA循环还依赖于另一种线粒体Fe-S蛋白——硫辛酸合成酶(LIAS)的功能。LIAS对于硫辛酸的合成以及重要硫辛酸盐基团的共轭都非常关键,硫辛酸盐基团由PDH (E2)和α-酮戊二酸脱氢酶(二氢硫辛酰胺S-琥珀酰转移酶,DLST)组成。LIAS对Fe-S蛋白生物合成的破坏特别敏感,因为其Fe-S簇在催化过程中被消耗掉,这就要求不断地进行蛋白交换。事实上,干扰NFS1和ISCU表达会大量减少PC9细胞中PDH-E2和DLST的脂化,然而干扰NNT对蛋白脂化无影响(图5E)。总的来说,这些数据表明,NNT引发酶和代谢缺陷,它们的缺陷与Fe-S蛋白生物合成的破坏相关,但NNT不可能直接影响这一过程。这反映出干扰NNT和ISCU后对TCA循环产生类似但又不同的影响。

6 NNT的缺失破坏脂肪酸代谢

除了消耗TCA循环中间体外,NNT缺陷细胞的LC-HRMS代谢组学分析表明脂肪酸代谢失调。干扰NNT促进长链脂肪酰基肉碱的大量积累,而长链脂肪酰基肉碱是脂肪酸β-氧化的底物(图6A)。鉴于在NNT缺陷细胞中存在呼吸系统缺陷,我们推测这些酰基肉碱的增加是由于脂肪酸氧化减少。我们发现在H1299和PC9细胞中OCR与棕榈酸盐的氧化有关,干扰NNT后OCR降低(图6B)。我们还观察到NNT缺陷细胞中饱和脂肪酸和不饱和脂肪酸均大量积累(图6C)。脂肪酸合成需要消耗大量NADPH,而且NNT干扰后NADPH的有效性降低,于是我们认为这些脂肪酸水平的增加是由于外源性脂肪酸摄入的增加。另外,我们发现,NNT缺陷细胞增强了对荧光棕榈酸类似物的吸收能力(图6D)。为了评价NNT干扰后脂肪酸的积累是否是一个潜在的不利因素,我们使用饱和脂肪酸棕榈酸酯刺激NNT缺陷细胞24小时,发现NNT干扰后使H1299和H2009细胞对棕榈酸盐敏感(图6E)。此外,NNT干扰后显著提高了H1299和PC9细胞对单一不饱和脂肪酸油酸盐的敏感性(图6F)。鉴于外源性脂肪酸补充的有害影响,我们预计细胞外脂质消耗将保护NNT的缺失。令人惊讶的是,培养基中的脂质消耗加剧了NNT的下调(图6G)。更重要的是,NNT干扰后细胞外脂质缺少会增加NADPH的消耗(图6H),这表明在缺乏外源性脂肪酸的情况下被迫合成脂肪酸,使NNT缺陷细胞中NADPH的有效性降低。总的来说,这些数据表明NNT可能在调节脂肪酸代谢中起重要作用,NSCLC细胞的脂肪酸代谢紊乱可能是一个可利用的弱点。

7 NNT失后线粒体靶向过氧化氢酶(MitoCatalase)挽救Fe-S蛋白功能

Fe-S蛋白对分子氧和更多有害物质的氧化非常敏感。虽然我们没有观察到线粒体蛋白抗氧化系统的氧化状态发生变化,但这并不排除NNT干扰后引发的线粒体ROS足够将这些敏感性辅因子氧化。为了探究这种可能性,我们使用了线粒体靶向的过氧化氢酶(MitoCatalase)来增强线粒体的抗氧化能力。我们成功地在H1299、H2009和PC9细胞中过表达了MitoCatalase (图7A)。MitoCatalase的表达也部分减弱了NNT干扰后相关线粒体中H2O2的产生(图7B)。这与NNT干扰后呼吸链复杂活动的减少相对应(图7C和7D)。另外,MitoCatalase的表达恢复NNT干扰后ACO2的活性(图7E)。综上所述,这些数据表明,提高线粒体解毒H2O2的能力可以避免NNT干扰后Fe-S蛋白发生缺陷,从而使NNT在避免Fe-S蛋白发生氧化方面发挥作用,但对Fe-S簇的生物合成无作用。

结论

总之,我们的研究表明,NNT对肺癌生物学具有重要意义,主要是通过调控Fe-S蛋白而促进线粒体代谢。与以往评价NNT功能的研究不同,我们描述了NNT对NSCLC中线粒体氧化还原稳态的微妙影响。在NSCLC中,NNT活性可能减轻了大量氧化代谢引起的氧化应激反应。我们的发现进一步表明了线粒体代谢在肺癌发生中的必要性,说明线粒体功能的增强可能是肺癌治疗的一个重要靶点。酶TPH-1对肾保护起关键作用,可能作为治疗CKD的潜在治疗靶点。
