深度解析医学证据,DeepEvidence为你支撑决策 背景:Janus激酶抑制剂(JAKi)是类风湿关节炎(RA)的有效治疗药物。口服监测研究(Oral Surveillance)以及近期 RABBIT 注册研究的数据均提示,RA 患者长期使用 JAKi 可能与癌症风险升高相关。巨噬细胞是肿瘤组织中浸润最丰富的免疫细胞亚群,肿瘤免疫监视依赖于巨噬细胞高效介导的吞噬作用以及对凋亡细胞的胞葬作用。巨噬细胞的极化状态和效应功能受到细胞因子依赖性 JAK-STAT 信号通路的严格调控,肿瘤相关巨噬细胞根据其活化状态可发挥抗肿瘤或免疫抑制功能。本研究假设 JAKi 可能将单核细胞来源的巨噬细胞重编程为免疫耐受状态,从而影响 RA 患者的肿瘤免疫监视机制。 目的:探讨 JAKi 对 RA 患者循环单核细胞亚群、巨噬细胞极化、细胞因子分泌以及与肿瘤免疫监视相关的巨噬细胞清除功能的影响。 方法:研究纳入健康供体(HD)、未接受 JAKi 治疗的 RA 患者以及接受 JAKi 治疗的 RA 患者,采集外周血分离单核细胞。采用流式细胞术分析循环单核细胞亚群和单核细胞来源的髓系抑制细胞(M-MDSCs)。
