Front Psychiatry:脑源性神经营养因子在抑郁症检测及监测中的应用

目前没有促进抑郁症检测并监测抗抑郁药效率的敏感和稳定的生物标志物。因此,本研究旨在调查脑源性神经营养因子(BDNF)的DNA甲基化和组蛋白修饰在监测针刺的严重程度和抗抑郁效应方面的潜力。通过社会隔离和慢性不可预见的轻度压力(CUMS)来建立抑郁症大鼠模型。采用酶联免疫吸附试验(ELISA)检测血清BDNF的表达,Western blot检测海马BDNF、组蛋白H3赖氨酸9(acH3K9)和HDAC

目前没有促进抑郁症检测并监测抗抑郁药效率的敏感和稳定的生物标志物。因此,本研究旨在调查脑源性神经营养因子(BDNF)的DNA甲基化和组蛋白修饰在监测针刺的严重程度和抗抑郁效应方面的潜力。

通过社会隔离和慢性不可预见的轻度压力(CUMS)来建立抑郁症大鼠模型。采用酶联免疫吸附试验(ELISA)检测血清BDNF的表达,Western blot检测海马BDNF、组蛋白H3赖氨酸9(acH3K9)和HDAC2的乙酰化水平,RT- PCR检测海马BDNF mRNA的表达。MS-PCR检测BDNF启动子I的DNA甲基化模式。

结果显示,与对照相比,血清和海马中BDNF的表达明显下调。在BDNF的mRNA中发现相同的趋势。值得注意的是,与模型组相比,针刺可逆转血清和海马BDNF的下调和BDNF mRNA的下调。针灸逆转CUMS诱导的海马acH3K9的下调。相反,CUMS诱导的海马HDAC2在模型组中的上调被针刺明显逆转。

总的来说,针刺的抗抑郁作用可能通过调节BDNF的DNA甲基化和组蛋白修饰来介导,BDNF或是用于检测抑郁症和监测严重程度和抗抑郁作用的新型生物标志物。
