Sirtuins是一类脱乙酰酶,可修饰结构蛋白,代谢酶和组蛋白,从而改变细胞蛋白的定位和功能。在哺乳动物中,有七种sirtuins参与与衰老、变性或癌症相关的氧化应激或代谢稳态等过程。研究人员通过qRT-PCR研究了sirtuins基因在不同不孕症诊断组的试管婴儿患者和卵母细胞捐献者(OD;对照组)的人类壁膜颗粒-黄质细胞(hGL)中的基因表达。

研究1:sirtuins基因在无卵巢因子女性中的表达水平及与年龄和IVF参数的相关性。结果发现,≥40岁的患者中,SIRT1、SIRT2和SIRT5的表达水平明显高于OD以及27-39岁、有输卵管、男性因素或无卵巢因素(NOF)的女性。仅SIRT2、SIRT5和SIRT7的表达与年龄相关。

 

研究2:sirtuin基因在女性不良反应者(PR)、子宫内膜异位症(EM)和多囊卵巢综合征中的表达。结果显示,与NOF对照相比,所有诊断组中的SIRT2基因表达均较高,而SIRT3、SIRT5、SIRT6和SIRT7的表达仅在PR中较高。与临床参数相关的SIRT1、SIRT6和SIRT7与EM患者的FSH和LH剂量呈正相关。PR中取回的成熟卵母细胞的数量与SIRT3、SIRT4和SIRT5的表达水平呈正相关。

综上所述,这些数据表明,PR卵泡中的细胞生理病理可能与DNA累积损伤有关,有必要行进一步的研究。
