Rheumatology:碱性磷酸钙晶体诱导肌腱细胞外基质重塑酶的表达

研究碱性磷酸钙晶体对肌腱细胞功能和活力的直接影响。

目的:碱性磷酸钙(BCP)晶体肌腱疾病相关的几种综合征有关,包括急性钙化性肌腱炎密尔沃基肩部综合征BCP 晶体和肌腱细胞之间的相互作用可能导致这些临床综合征。该研究旨在确定 BCP 晶体对肌腱细胞功能和活力的直接影响。

方法:采用体外实验评估用BCP晶体培养的人肌腱细胞的变化。实时PCR用于确定肌腱相关基因和细胞外基质重塑酶(金属蛋白酶,MMPs;去整合素和金属蛋白酶,ADAMTS;金属蛋白酶组织抑制剂,TIMPs)表达的变化。ELISA用于测量肌腱细胞上清液中的蛋白质浓度。MTT和alamarBlue™ 实验用于确定细胞活力的变化。

结果:24小时后,BCP 晶体上调了MMP-1、MMP-3、ADAMTS-4和TIMP-1的肌腱细胞基因表达。时间进程实验表明,TIMP-1表达在8小时达到峰值、MMP-1和ADAMTS-4表达在48 小时达到峰值。48小时后,环氧化酶-1(COX-1)基因表达上调。

 肌腱细胞不改变巩膜和肌腱胶原的表达,BCP晶体不诱导促炎细胞因子的表达。24小时后,BCP晶体增加肌腱细胞释放前列腺素E2(PGE2)和MMP-1蛋白 。然而,COX-1抑制剂和COX-2抑制剂均未导致BCP晶体诱导的肌腱细胞中细胞外基质重塑酶基因表达的一致变化。BCP晶体对肌腱细胞活力没有影响。

结论:BCP晶体在肌腱细胞中诱导细胞外基质重塑酶,但不诱导炎性细胞因子
