大多数的CRC起源于肿瘤前的前体病变:结肠息肉。息肉切除术可防止结肠息肉发展为CRC。筛查时发现的息肉的数量和大小也与患者未来罹患CRC的风险相关。尽管如此,目前尚无相关化学预防措施可以降低患者发生进一步息肉或CRC的风险。


在该研究中,研究人员旨在研究5-ASA(5-氨基水杨酸,一种治疗溃疡性结肠炎的药物)作为预防散发性CRC药物的潜力。

研究人员通过使用5-ASA处理人结肠直肠腺瘤、转化的腺瘤和癌细胞系,探究在二维和三维(3D)培养中细胞的生长状况,β-catenin的转录活性以及对肿瘤细胞干细胞特性的影响。


结果显示,5-ASA可抑制腺瘤细胞的生长并抑制β-catenin的转录活性。在3D培养模型中,研究人员发现β-catenin的下调可抑制干细胞标记物LGR5的表达水平,并在功能上抑制人腺瘤和癌细胞的干细胞特性。

5-ASA降低了PC/AA/C1腺瘤和LS174T癌细胞的肿瘤球形成能力

总而言之,该研究结果显示,5-ASA可以抑制腺瘤衍生细胞的干细胞特性。研究人员表示,价格合理且耐受性良好的5-ASA是化学药物预防的一个潜在的候选药物,其可降低高风险人群中结直肠息肉和CRC的风险。
