Diabetologia:幼年发展为β细胞自身免疫的儿童早期DNA甲基化变化

     1型糖尿病是一种病因复杂的慢性自身免疫性疾病,包括潜在的表观遗传调节作用。以往的表观基因组学研究主要集中在临床诊断的个体上。这项研究的目的是评估与1型糖尿病相关的早期DNA甲基化变化,这些变化在诊断之前或甚至在自身抗体出现之前就已经存在。

     方法:采用简化代表性亚硫酸氢盐测序法,对7例1型糖尿病自身抗体阳性者和年龄、性别、风险、出生地匹配的对照组2 2 6例外周血单个核细胞进行纯化的CD4+T细胞、CD8+T细胞和CD4HLACD8DNA甲基化水平的研究。结果7例1型糖尿病自身抗体阳性者和2 2 6例正常对照外周血单个核细胞中CD4+T细胞、CD8+T细胞和CD4−CD8−细胞组分的DNA甲基化水平均无显著性差异(P>0.0 5)。我们还利用同一样本的RNA测序数据探索了DNA甲基化和基因表达之间的相关性。使用焦磷酸测序对RRBS结果进行技术验证。

     结果:在1型糖尿病自身抗体阳性个体和对照组之间,我们分别在CD4+T细胞、CD8+T细胞和CD4−CD8−细胞组分中发现了79、56和45个差异甲基化区域。血清转换前样本的分析确定了疾病早期的DNA甲基化特征,包括CD4+T细胞中IRF5启动子的差异甲基化。此外,我们还利用以下cpG位点的焦磷酸测序验证了rrbs的结果:在ARRDC2启动子上的chr19:18118304;在pcBP3内含子上的chr21:47307815;以及在cd4+T c e l l s中靠近traf3的基因间隔区的chr14:81128398。

    结论:这些初步结果为基因的细胞类型特异性差异表观遗传调控提供了新的见解,这可能有助于1型糖尿病在疾病发展的非常早期阶段的发病机制。如果这些发现得到证实,它们可能成为对疾病预测和管理有用的潜在标志。
