镰状细胞血症L-谷氨酰胺氧化应激

研究认为药用级L-谷氨酰胺可降低镰状细胞血症患者疼痛危象发生率。

氧化应激在镰状细胞病的病理生理过程中发挥了关键作用。药物级L-谷氨酰胺被证明可增加镰状红细胞中还原型烟酰胺腺嘌呤二核苷酸的比例，减少氧化应激，改善镰状细胞相关疼痛的发生。

近期，研究人员开展III期临床研究，考察口服0.3g/Kg体重药物级L-谷氨酰胺对镰状细胞性贫血或镰刀型β0-地中海贫血患者48周疼痛危象发生率的影响。

230名患者参与研究，年龄范围5-58岁，女性占53.9%，其中152人接受L-谷氨酰胺，78人接受安慰剂。治疗组疼痛危象发生率降低，48周谷氨酰胺组的危象平均发生2.0次，安慰剂组为3.0次。2/3的患者在研究期间坚持服用羟基脲。谷氨酰胺组低度恶心、非心脏性胸痛、疲劳和肌肉骨骼疼痛率较安慰剂组增加。

研究认为药用级L-谷氨酰胺可降低镰状细胞血症患者疼痛危象发生率。
