脂肪细胞膜蛋白:治疗前列腺癌新靶点

前列腺癌是一种泌尿系统恶性肿瘤,对中老年男性造成了巨大的危害。目前,国际公认的研究表明,通过抑制雄性激素的合成,对于治疗前列腺癌已经取得了较好效果。但是,相当一部分患者会逐渐产生抗药性并最终导致癌细胞转移。

图  高胆固醇促发前列腺癌的可能机制

近日,中国科学院苏州生物医学工程技术研究所(以下简称中科院苏州医工所)肿瘤标志物中心高山实验室在国际权威期刊Cancer Research发表题为“Cholesterol Induces Epithelial-to-Mesenchymal Transition of Prostate Cancer Cells by Suppressing Degradation of EGFR through APMAP”的研究论文,首次揭示了脂肪细胞相关膜蛋白(APMAP)可作为前列腺癌诊断和治疗的标志物,并阐述了其背后的分子机制。

研究者们从高胆固醇会促发前列腺癌这一现象入手,通过使用胆固醇刺激体外培养的前列腺癌细胞,将癌细胞打碎,提取细胞膜上的亚细胞组分进行检测,发现了两类靶向蛋白,分别是表皮生长因子受体(EGFR)以及脂肪细胞相关膜蛋白,高胆固醇能够诱导这两种蛋白的表达,从而促进前列腺癌症的发展。该研究发现为前列腺癌患者的治疗提供了新靶点,针对该靶点研发出的新药物,有望进一步降低前列腺癌症的发展以及侵袭转移。
