Clinica Chimica Acta:肝素干扰检测风疹IgM的机制

肝素可能通过与风疹病毒抗原和磁性颗粒表面包被的抗人IgM特异性抗体结合而参与反应,从而影响检测结果。

本研究的目的是探讨肝素在检测LIAISON®风疹IgM(Rub-M)中的作用以及干扰机理。研究人员将不同浓度的肝素锂和肝素钠添加到十份血清样品中。使用LIAISON XL检测系统测量Rub-M的相对光单位(RLU)值。将不同水平的IgM血清与磁性颗粒在Rub-M检测试剂盒中于4°C孵育4小时,以封闭包被在磁性颗粒表面的抗人IgM特异性抗体。另外,将Rub-M检测试剂盒中的风疹病毒抗原替换为磷酸盐缓冲液(PBS)。经过上述两种不同的处理后,测量LIAISON®Rub-M原始血清和含有各种浓度肝素的血清的RLU值。

研究结果显示,LIAISON®Rub-M的RLU值随肝素含量的增加(低于40 IU / mL)而增加,并在40–50 IU / mL时达到峰值。随后,LIAISON®Rub-M的RLU值随肝素浓度的降低而降低。当风疹病毒抗原被PBS替代时,含有40 IU / mL肝素的血清样品的LIAISON®Rub-M的RLU值显着降低。 IgM的阻断浓度逐渐增加,七个包含40 IU / mL肝素的血清样品的LIAISON®Rub-M的RLU值也逐渐降低。

研究结果表明,含肝素的血浆不能用于检测LIAISON®Rub-M。肝素可能通过与风疹病毒抗原和磁性颗粒表面包被的抗人IgM特异性抗体结合而参与反应,从而影响检测结果。
