近日,国际杂志 《Clinical Chemistry》上在线发表一项关于检测肽标记的细菌性呼吸道病原体的基因组学方法的研究。

快速鉴定呼吸道病原体可能有助于靶向抗微生物治疗。通过基质辅助激光解吸/电离飞行时间质谱法直接鉴定支气管肺泡灌洗液(BAL)中的细菌会被干扰物质所混淆。研究人员通过对5种革兰氏阴性细菌的独特肽标记物,使用液相色谱-串联质谱法(LC-MS/MS)的方法鉴定BAL中的病原体。

在分别对代表鲍氏不动杆菌,卡他莫拉菌,铜绿假单胞菌,嗜麦芽窄食单胞菌和肺炎克雷伯杆菌的14-25个全基因组进行翻译和消化,并确认每个构成理论核心肽结构。进行快速胰蛋白酶消化;肽通过LC-MS/MS分析,并与理论核心肽进行比较。用最近共同祖先搜索来鉴定和分析高可信性核心肽(假发现率<1%)以产生潜在的物种特异性肽标记。用蛋白质BLAST验证了每种肽的物种特异性。此外,将12个病原体连续稀释成合并发炎的BAL,并使用目标LC-MS / MS测定法同时检测25种肽。

每种病原体丰富度最高的五种独特肽区分这些病原体具有不同的检测灵敏度。有肽标记物鲍曼不动杆菌和铜绿假单胞菌同时进入发炎的BAL时,用目标LC- MS / MS分别可以检测到至少3.60.2)×1032.20.6)×103菌落形成单位,。

这种验证研究显示了在BAL中鉴定5种革兰氏阴性细菌病原体的特异性肽段的可行性,并且可以提供一种用于快速直接鉴定BAL中细菌病原体的新方法。
