Cell Res: 重大进展!同济大学陈义汉院士/梁丹丹发现心肌细胞增殖的关键调节蛋白,有望成为心脏再生的干预目标

许多心脏病与心肌细胞的丢失或功能障碍有关。然而,成年心肌细胞在新生儿阶段狭窄的增殖窗口后会大大丧失细胞分裂和增殖的能力。作为代表性的器官,再生能力差,成年心脏特别容易受到损伤反应的破坏。

考虑到心肌细胞的有丝分裂后性质,成年哺乳动物心脏被认为是终末分化的器官。因此,通过心肌细胞增殖进行心脏修复的潜力非常有限。低密度脂蛋白受体相关蛋白6(LRP6)是Wnt共同受体,是胚胎心脏发育所必需的。

2020年9月24日,同济大学陈义汉院士及梁丹丹共同通讯在Cell Research在线发表题为“LRP6 downregulation promotes cardiomyocyte proliferation and heart regeneration”的研究论文,该研究发现LRP6通过调节心肌细胞增殖在心脏修复中的作用。

Lrp6缺失增加了新生,幼年和成年小鼠的心肌细胞周期活性。小鼠心脏中Lrp6的心肌细胞特异性缺失在心肌梗死(MI)后引起强烈的再生反应,导致MI面积减少和左心收缩功能改善。体内遗传谱系追踪显示,MI后Lrp6缺陷小鼠心脏中新形成的心肌细胞主要来自常驻心肌细胞。此外,该研究发现Lrp6缺乏的促增殖作用是由ING5 / P21信号通路介导的。使用腺相关病毒(AAV)9 miRNAi-Lrp6构建体的基因治疗促进了小鼠心脏损伤的修复。Lrp6缺乏症还诱导了人类诱导的多能干细胞衍生的心肌细胞(iPSC-CMs)的增殖。该研究确定LRP6是心肌细胞增殖的关键调节剂,这可能导致开发一种新型分子策略来促进心肌的再生和修复。

哺乳动物组织表现出特定的速率和细胞更新机制,不同器官的修复能力差异很大。新生小鼠和成年斑马鱼的谱系图分析表明,大多数再生的心肌细胞都来自于既存的心肌细胞。此外,许多研究表明,成年哺乳动物心脏能够在受伤后心肌细胞可以增殖,这表明心肌细胞增殖可能是修复心脏的有效策略。因此,了解心肌细胞增殖能力的机制以及如何控制心肌细胞增殖过程是成功修复心脏损伤的关键。

低密度脂蛋白受体相关蛋白6(LRP6)是一种单跨膜蛋白,在经典Wnt /β-catenin信号传导中起关键作用,已被证明对心脏神经系统至关重要。小鼠Lrp6的消融影响心脏祖细胞的增殖和存活,并在早期心脏发生过程中造成一系列严重的形态发生缺陷。成年后,LRP6功能障碍与冠心病和致命性心律失常有关。关于LRP6在心肌细胞周期活动中的潜在作用知之甚少。

在该研究中,发现Lrp6缺乏症会在小鼠心脏和人类诱导的多能干细胞衍生的心肌细胞(iPSC-CMs)的不同阶段增加心肌细胞的增殖。小鼠心脏中的Lrp6缺乏症减少了心肌梗塞(MI)的大小并改善了心脏功能。ING5 / P21信号通路被证明负责LRP6对心肌细胞增殖的影响。损伤后,施用编码miRNAi-Lrp6的AAV9构建体可促进小鼠的心脏修复。因此,该研究揭示了一种控制心肌细胞周期再进入的新型分子开关,并有望成为心脏再生的干预目标。
