INTREPID研究是一项为期12周、随机、双盲、安慰剂对照的II期研究,评估了EDP-305治疗原发性胆源性胆管炎(PBC)的安全性、耐受性、药代动力学和疗效。该研究的主要终点是评估在第12周时碱性磷酸酶(ALP)较治疗前值降低至少20%的受试者比例(ALP响应)。

在意向性治疗(ITT)分析中,与安慰剂组的11%(n = 1/9)相比,EDP-305 1 mg和2.5 mg治疗组产生了45%(n = 14/31,p = 0.106)和46%(n = 13/28,p = 0.063)的ALP响应。与安慰剂组相比,EDP-305 1 mg组(p = 0.017)和2.5 mg组(p = 0.021)在12周时ALP绝对变化具有统计学意义。

在ITT分析中,关键的次要终点(包括丙氨酸氨基转移酶、天冬氨酸氨基转移酶和γ-谷氨酰转移酶相对于基线的变化)与安慰剂相比在统计学上也有显著差异。
