来自EPFL的研究人员现在就表示朝着这一目标迈出了新的一步--人类对一种水果衍生化合物的临床试验显示,这种化合物有望减缓老年患者的线粒体衰老并且目前还没有发现任何副作用。

据了解,一种名为尿黄素A (以下简称UA)的化合物是这项研究的重点,此前的实验表明,它有望延长蠕虫和老鼠的寿命。这种化合物似乎可以通过改善线粒体的功能来对抗衰老,其作用方式跟运动到来的好处类似。虽然它不存在于任何已知的食物中,但石榴和覆盆子等水果中的生物分子则能在人体肠道中分解成UA。

在新的这项研究中,研究小组合成了UA化合物,并以不同剂量让60名老年参与者服用。这些人日常习惯久坐不动,但在实验期间他们的身体却表现健康。研究小组测试的目的则是为了确定这种化合物的安全性以及它所带来的功效。

在第一轮测试中,研究人员给一些参与者单次口服剂量在250到2000毫克之间的UA。跟对照组相比,实验组没有发现任何副作用。接下来,研究小组在28天的时间里测试了这种化合物的安全性和有效性。研究对象被分成四组,每组每天接受不同的UA口服剂量:250毫克、500毫克、1000毫克以及对照组接受安慰剂。

同样,这些参与者在长期服用后都没有表现出任何得副作用。研究人员从患者的血浆样本中检测出了UA的生物标志物,发现它们在所有剂量下都存在。重要的是,研究小组发现,当剂量分别为500毫克和1000毫克时,骨骼肌细胞中线粒体的基因表达受到影响。

该研究的作者之一Johan Auwerx表示:“这些建立在以往临床前试验基础上的最新发现,确实明确了UA是如何可能成为人类健康的游戏规则改变者。”

EPFL一家子公司Amazentis正在计划尽快将UA产品推向市场,但毫无疑问,在这之前它还需要做进一步的工作。

关于这项新研究的报告已发表在《Nature Metabolism》上。
