确认这些异种肿瘤细胞缺失在小鼠体内再现原肿瘤后,研究人员对这些肿瘤细胞进行2D及3D培养并测试某些药物组合的疗效以及肿瘤是否会对药物产生耐药性。药物处理后研究人员对细胞进行染色从而简单判断细胞存活情况,以明确药物疗效,并据此将药物粗略地分为能够阻止肿瘤生长和对肿瘤无效果两大类。

研究人员发现三阴乳腺癌细胞对某些类别的药物敏感,例如影响细胞内部和周围物理因素的药物,但对其他药物具有耐药性。研究人员指出,药物反应性方面,2D培养(细胞单层)的细胞与3D培养存在差别,3D培养的细胞结构特征更复杂,与真实肿瘤中更为相似。虽然本研究只对一种肿瘤进行了造模研究,测试的也仅仅是一些临床常见药物,但研究中用到模型体系和简单便捷的筛选方法可以推广到所有肿瘤及目前市面上已批准的任何药物。我们还可以将这种实验方法进一步拓展,用以寻找有效方法来治疗那些过去我们束手无策的罕见高侵袭性肿瘤。
