CELL:还敢熬夜吗?缺觉使得肠道积累活性氧而致死

睡眠对生存至关重要,睡眠行为无处不在,在最早的动物中明显存在类似睡眠的状态,而且严重的睡眠缺失可能是致命的。但是至今为止,这种致命性的原因尚不清楚。

最近,研究人员使用苍蝇和小鼠模型发现,睡眠剥夺能够导致活性氧物种(ROS)的积累和随之而来的氧化应激,特别是在肠道。

ROS不仅是睡眠剥夺的相关因素,而且是死亡的驱动因素:ROS的中和可以防止氧化应激,并使苍蝇在很少或没有睡眠的情况下有一个正常的寿命。

因此,我们可以通过口服抗氧化剂化合物或肠道靶向转基因表达的抗氧化酶来抵抗睡眠不足的危害。

因此,研究人员得出的结论是,严重睡眠限制后的死亡可能是由氧化应激引起的,肠道是这个过程中的中心,当防止ROS积累时,没有睡眠的生存是可能的。

《Anaesthesia 》上一项研究将睡眠不足与DNA损伤联系在了一起。研究人员在不同时间点提取了49名健康的全职医生的血液并进行分析,结果显示被要求在现场通宵工作的医生比未通宵工作的医生DNA修复基因表达度更低,DNA断裂更多。这些需要通宵工作医生仅仅经历了一个晚上的睡眠剥夺,体内DNA的修复基因表达度便有所减少,受损DNA也随之增加!
