已有研究通过不同的模型对衰老过程中蛋白质稳定性丧失进行了很好的描述,但关于衰老过程中生物体液中蛋白质聚集水平的研究却仍很少。因此,本研究的目的是使用两种不同的方法:利用构象特异性抗体和FTIR光谱分析来评估与年龄相关的蛋白质在人血浆中的聚集模式。其中,FTIR光谱分析已被广泛应用于生物医学研究中,研究蛋白质在健康和疾病中的构象变化。

研究人员先采用位点印迹分析对来自葡萄牙阿威鲁地区的初级保健基础队列的样本进行分析,随后使用构象特异性抗体进行免疫检测,并获取FTIR光谱。免疫印迹分析显示,随着年龄的增长,人血浆中的蛋白质构象原纤化前低聚物逐渐减少和原纤化结构增加。这一发现也得到了PLS-R多变量分析FTIR数据的支持,其中可以观察到供体年龄与血浆蛋白二级结构之间的正相关关系。

研究表明,来自较年轻供体的样品的特征是具有反平行的β-折叠结构,而分子间β-折叠则具有较老的样品特征。 排除与年龄相关的合并症可改善蛋白质构象特征与衰老之间的相关性。结果揭示了中老年人血浆蛋白的结构变化,证实了蛋白质聚集随年龄的变化,支持了FTIR作为研究老年期蛋白质稳态的可靠方法的适用性。
