近期,北京生命科学研究所张志远实验室在《Chemical Communications》杂志上发表了题为“Activity-based Protein Profiling Reveals GSTO1 as the Covalent Target of Piperlongumine and a Promising Target for Combination Therapy for Cancer”的研究论文。该文章揭示了天然产物荜茇酰胺(piperlongumine)通过共价结合抑制GSTO1蛋白活性达到特异性杀伤癌细胞的作用,并发现GSTO1可作为癌症联合用药的有效靶点。

荜茇酰胺是一种分离于胡椒科植物荜茇(Piper longum)根部的生物碱类化合物。对荜茇酰胺研究表明其有多种药理学作用,其中最为引人注目的是其对肿瘤细胞的选择性杀伤作用,这使得荜茇酰胺近年来成为在肿瘤治疗方面极具发展潜力的化合物。然而,目前为止,荜茇酰胺在特异性杀伤癌细胞作用中的靶点及机理仍然存在很大争议。

作者首先通过一定的构效关系分析及模型反应确定了荜茇酰胺通过共价结合其靶点的方式产生特异性杀伤癌细胞的活性。作者根据构效关系信息设计了分子探针PL-5,通过基于点击化学的Activity-based Protein Profiling(ABPP)技术进行了靶标蛋白的鉴定,并确认荜茇酰胺与Glutathione S-transferase omega-1(GSTO1)的酶活催化位点Cys32发生共价结合。随后,进一步通过生化手段确认了荜茇酰胺对GSTO1的活性抑制作用以及GSTO1蛋白对于维持癌细胞活性的重要功能,并确认了荜茇酰胺特异性杀伤癌细胞的功能是通过抑制GSTO1的活性进行的。此外,通过将荜茇酰胺作为GSTO1抑制剂进行联合用药筛选,该研究还发现抑制GSTO1可以与大量的抗癌药物尤其PI3K/Akt/mTOR通路的抑制剂是产生明显的协同作用,揭示了GSTO1具有作为癌症治疗的药物联用靶点的巨大潜能。
