先前的研究表明,重组腺相关病毒(AAV)介导的下丘脑BDNF基因转移减轻了饮食诱导和遗传模型中的肥胖,糖尿病代谢综合征

最近,研究人员检测了内置自动调节系统的功效和安全性,以控制模仿中年小鼠身体自然反馈系统的转基因BDNF表达。用自身调节的BDNF载体或黄色荧光蛋白(YFP)对照处理12个月大的小鼠,维持正常饮食,并监测28周。 

BDNF基因转移阻止了与衰老相关的代谢下降的发展,其特征在于:预防与衰老相关的体重增加,减少肥胖,逆转棕色脂肪活性的下降,增加脂联素,同时减少循环中的瘦素和胰岛素,改善葡萄糖耐量,增加能量消耗,减轻肝脏脂肪变性,抑制下丘脑和脂肪组织中的炎症基因。此外,BDNF治疗减少了类似焦虑和抑郁的行为。

这些安全性和有效性数据提供了下丘脑BDNF是促进健康老化的靶标的证据。
