科学家发现一种“双管齐下”治疗非小细胞肺癌的新方法,通过既不让癌细胞吃东西也不让它分解自身蛋白质的方法来“饿死”它。

科学家发现一种“双管齐下”治疗非小细胞肺癌的新方法,通过既不让癌细胞吃东西也不让它分解自身蛋白质的方法来“饿死”它。

非小细胞肺癌是最常见的肺癌类型,而不少非小细胞肺癌是由KRAS基因突变导致。美国波士顿儿童医院等机构研究人员2日在美国《国家科学院学报》上发表论文说,胰岛素和胰岛素样生长因子-1是影响细胞生长的重要因素,他们以KRAS突变小鼠为研究对象,并设法完全阻断胰岛素和胰岛素样生长因子-1的信号传导,结果,小鼠肺部的肿瘤生长受到明显抑制。

不过,阻断胰岛素和胰岛素样生长因子-1的信号传导并不能完全抑制肿瘤生长。研究人员介绍,胰岛素样生长因子-1等生长因子会告诉细胞,周围有营养物质;抑制相关信号传导后,肿瘤细胞就会因无法摄取必需的氨基酸而“挨饿”。然而,肿瘤细胞并不会坐以待毙,而是会分解自身蛋白质来产生氨基酸。

研究人员由此开展动物实验,同时抑制胰岛素和胰岛素样生长因子-1的信号传导以及蛋白质的分解,结果发现,肿瘤生长几乎被完全抑制。
