安进公司新型肺癌药物motesanib III期研究结果令人失望

2012年4月1日，安进（Amgen）公司肺癌药物motesanib在III期研究中没有达到预期主要目标。 该药由安进公司与武田（Takeda）制药旗下Millennium公司联合开发。在III期研究中，motesanib结合2种化疗药物，未能延长晚期非鳞状非小细胞肺癌（advanced non-squamous non-small cell lung cancer，NSCLC）患者总体存活时间

2012年4月1日，安进（Amgen）公司肺癌药物motesanib在III期研究中没有达到预期主要目标。

该药由安进公司与武田（Takeda）制药旗下Millennium公司联合开发。在III期研究中，motesanib结合2种化疗药物，未能延长晚期非鳞状非小细胞肺癌（advanced non-squamous non-small cell lung cancer，NSCLC）患者总体存活时间。

安进公司研发副总裁Roger Perlmutter称："我们对此次试验的结果感到失望，但仍期待着进一步的数据分析，希望这些数据最终有助于今后该领域的研究。"

此次III期研究还有一些次要目标，包括无进展生存期，但这些数据尚未公布。安进公司表示，将在即将到来的医学大会上公布研究的详细结果。

Motesanib是一种非常新颖的候选药物，结合拮抗剂能对付NSCLC中的3种突变，同时也是这类药物中首个进入III期研究的药物。

Motesanib是一种口服小分子药物，靶向于血管内皮因子受体（vascular endothelial growth factor receptors ，VEGFR）-1、-2、-3和血小板衍生性生长因子受体，以及干细胞因子受体。

目前NSCLC的标准治疗药物包括阿斯利康（AstraZeneca）公司的Iressa和罗氏（Roche）公司的Tarceva，这2种药物均靶向于血管内皮生长因子受体。

在所有癌症类型中，NSCLC的死亡率最高。近期针对这种疾病所开展的临床试验屡屡失败，均未提供一种新的治疗药物，包括赛诺菲-安万特（Sanofi-Aventis）公司的aflibercept、诺华（Novartis）公司的ASA404及辉瑞（Pfizer）公司的figitumumab。

但这也不完全是坏消息。辉瑞（Pfizer）公司去年在美国癌症大会上提交的ALK基因抑制剂crizotinib II期研究结果令人印象深刻。

在今年1月，crizotinib被FDA授予"rolling"快车通道地位（'rolling' fast-track status），可能在今年早些时期能够上市。（生物谷bioon.com）
