研究背景:虽然大多数非酒精性脂肪肝的患者(NAFLD)只有脂肪变性,但有相当一部分人发展为非酒精性脂肪肝(NASH),可导致肝硬化和肝细胞癌(HCC)。许多已建立的NASH小鼠模型需要24-52周,这使得对药物反应的测试花费昂贵且耗时。

研究方法:研究试图通过西方饮食(WD)建立一种快速发展为广泛纤维化和HCC的NASH模型。西方饮食包括:高脂肪、高糖和高胆固醇联合低浓度的注射CCl4(每周腹腔注射),CCl4作为模型形成的加速器。

研究结果:C57BL/6J小鼠正常饮食±CCl4或西方饮食(WD)± CCl4。这种饲养方式进行12周或24周。CCl4的添加加重了WD引起的NASH、纤维化和肿瘤的进程,导致在12周出现3期纤维化和24周出现HCC。此外,肝脏的转录组学分析表明,WD/CCl4小鼠的分子通路失调和免疫学特征与人类NASH相似。
