该构建体设计有互连的隔室以容纳干细胞和内皮细胞。将细胞包封在优化的结冷胶(GG)基水凝胶基质中,使用锶(Sr2 +)离子交联以利用其生物活性,最后在不同大小的隔室内组装。钙(Ca2 +)-交联凝胶也用作比较Sr2 +成骨作用的对照组。

结果显示,Sr 2+离子在水凝胶基质内成功扩散并且在压缩载荷下增加水凝胶基质的强度。人TERT间充质干细胞(TERT-hMSCs)和人脐静脉内皮细胞(HUVECs)的体外共培养,包裹在含有GG-水凝胶的Sr2 +离子中,并在成骨条件下通过分隔支架相互连接,增强细胞活力。并且支持骨生成,碱性磷酸酶活性显着增加,骨桥蛋白和骨钙素与Ca2 +交联的GG-PCL支架相关。

综上所述,这些结果表明,TERT-hMSCs和HUVEC区室化共培养物的设计和制造使得能够有效地研究和促进骨生成。
