8月14日发表在欧洲心脏杂志上一项研究表明,年轻的心肌球来源细胞可通过心脏基因表达、心脏功能、运动能力和系统生物标记物等方法恢复衰老动物的活力。我们离“干掉”衰老或许又近了一步。

希达-西奈心脏研究所(Cedars-Sinai Heart Institute)的最新一项研究表明,心脏干细胞(CSC)的输注或有助于逆转心脏衰老过程,使年老心脏更年轻。研究结果于当地时间8月14日发表在欧洲心脏杂志上《European Heart Journal》。

希达-西奈心脏研究所主任、也是这项研究的主要研究人员Eduardo Marbán博士表示,“我们先前的实验室研究和人体临床试验已证明,心脏干细胞输注治疗心力衰竭的前景。现在我们发现这些干细胞可以逆转与心脏老化有关的问题。”

研究人员将新生的实验室大鼠的心肌球来源细胞(cardiosphere-derived cell,CDC)注射入平均年龄为22个月的大鼠心脏中,22个月的大鼠已经算是老年鼠。对照组则接受安慰剂治疗,用生理盐水(phosphate-buffered saline, PBS)注射代替干细胞。然后将两组老年大鼠与平均年龄为4个月的年轻大鼠进行比较。

随后通过超声心动图、跑步机压力测试和血液分析测试所有大鼠的心脏功能。接受年轻大鼠的心肌球来源细胞注射的老年大鼠一个月后再进行一轮测试。

Marbán博士表示,“细胞逆转衰老的方式很吸引人。它们分泌细小的囊泡,这些小囊泡充满了RNA和蛋白质等信号分子。年轻细胞的囊泡似乎包含了逆转衰老有需要的所有指令。”

注射了心肌球来源细胞的大鼠与对照组相比,变化如下:

·心脏功能得到很大的改善

·端粒酶的活性增加,端粒长度延长

·锻炼能力提高了20%,体重减少了30%

·毛发再生能力更强

这些结果表明,年轻的心肌球来源细胞可通过心脏基因表达、心脏功能、运动能力和系统生物标记物等方法恢复衰老动物的活力。

研究第一作者Lilian Grigorian-Shamagian博士表示,“这项研究还没有评估接受心肌球来源细胞是否会延长寿命,所以我们还需要做更多的研究。包括研究具有逆转衰老功能的心肌球来源细胞是否需要来自于年轻的捐赠者;细胞外的囊泡是否可以有同样的效果等。

自2009年,Marbán和他的研究团队完成了世界上第一次用病人自己的心脏组织来培养特殊的心脏干细胞。然后,这种干细胞被注射回病人心脏中来努力修补心脏病发作导致的心脏受损和再生出健康心肌。随后,Marbán团队研究使用干细胞治疗杜氏肌营养不良症患者以及患有心力衰竭的患者。
