短肠综合征是一种损失部分小肠的疾病,会降低患者的营养吸收能力。目前的疗法是移植,但是可移植的器官数量有限,而且由于植入失败和细胞排斥问题,移植器官的三年存活率较低。使用源自患者的细胞制造再生肠进行移植,或可以解决这些问题,不过之前是使用合成支架构建这些器官,成功率有限。去细胞化肠一直被认为是一种有前景的替代性支架,但是截至目前,利用这种支架所得的器官缺乏恢复营养吸收能力所需的功能性血管。 Harald Ott 及同事制造出的一种支架既能实现血管化,又能实现营养传输。

他们采用一种可以保留支架血管的方法,移除了一节大鼠肠中的细胞,之后为了恢复营养吸收能力,再增加两种肠细胞。首先,作者使用人类干细胞源上皮细胞制造出多重细胞球体,被称为迷你肠道球体,它们会借助一个硅胶管在支架上融合起来。经过两周的培养后,再添加内皮细胞。结果,营养传输率与大鼠原生肠样本的传输率相仿,而且在植入大鼠四周之后,这种生物工程肠继续存活并不断成熟。

本研究的主要影响体现在制造出了一种支架,它可以充分血管化,并且能够使内皮肠细胞再生,就像在健康肠道内一样,从而使移植肠的存活期更长。虽然该方法还没有在人体身上进行测试,而且据此制造出来的肠管也没有完全达到原生肠的成熟度,但是在大小和营养吸收能力方面已取得了一定的技术进步,这意味着生物工程制造的功能性移植肠未来前景可观。
