研究人员采用无IgG μ重链的成熟B细胞缺陷的小鼠和输注IgG来研究IgG的功能。采用高脂饮食(HFD)诱导肥胖。用Sambucus nigra凝集素印迹法评估小鼠IgG蛋白Fc多糖的相对唾液酸化(进而影响IgG Fc受体激活)。用唾液酸前体N-乙酰-D-甘露胺(ManNAc)处理HFD喂养的小鼠来检测正常化IgG唾液酸化的效应。

B细胞缺陷小鼠不容易罹患肥胖诱导性高血压。与来自对照chow喂养的小鼠的IgG相比,来源于HFD喂养的小鼠的IgG低唾液酸化,将其输注给缺乏IgG的受体小鼠时,可升高受体小鼠的BP;如果受体小鼠缺乏FcγRIIB,则无上述高血压反应。神经氨酸苷酶处理的IgG,缺乏Fc多糖末端的唾液酸,也可升高血压。在培养的内皮细胞中,通过FcγRIIB、来源于HFD喂养的小鼠的IgG和神经氨酸苷酶处理的IgG改变内皮细胞NO合成酶的磷酸化来抑制血管内皮生长因子激活内皮细胞NO合成酶。在人类中,肥胖与低IgG唾液酸化由关,收缩压与IgG唾液酸化逆相关。内皮细胞FcγRIIB缺乏的小鼠不容易罹患肥胖诱导性高血压。此外,对于HFD喂养的小鼠,ManNAc可正常化IgG唾液酸化,预防肥胖诱导性高血压的发生。

在小鼠中,内皮细胞中的低唾液酸化的IgG和FcγRIIB在肥胖诱导性高血压的发生过程中具有至关重要的作用,而且在人类中获得了相应的支持证据。靶向这些机制的干预措施,如补充ManNAc,或可打断肥胖和高血压之间的联系,从而预防肥胖诱导性高血压的发生。
