AGING CELL:长期饮食限制使造血干细胞年轻化

造血细胞移植(HCT)是治疗多种造血恶性肿瘤的关键方法。随着世界老龄化人口显著扩大,高龄HCT接受者和供者增加。然而,捐献者年龄超过60岁后每递增10年,HCT后总死亡率的危险率就会增加5.5%。

研究发现,老年HSCs会表现出重建衰退,尤其是在淋巴再生方面。因此,探索提高来自老年供体的HSCs的再造能力的方法可能对HCT有相当大的意义。

此前,研究人员发现,早期的饮食限制(DR)可以改善早期衰老的C57BL/6 J小鼠的HSC的再生能力。然而,到目前为止,中期DR对临床HCT中主要使用的分选HSCs和未纯化的造血细胞的影响还没有报道。

最近,来自南昌大学第二附属医院的研究团队在Aging Cell杂志发文,其分析了从15-18个月龄开始,应用30%的DR方案短期(4个月)和长期(9个月)处理C57BL/6 J小鼠后,对移植中以及未受干扰条件下的HSCs和骨髓细胞的功能的影响。15-18个月龄的小鼠等效于人类的大致50-60岁。

研究人员发现,短期DR显著降低了老龄小鼠HSC库的大小,尤其是骨髓型HSCs(CD150highHSCs和CD41+HSCs),而淋巴型HSCs(CD150lowHSCs和CD41-HSCs)没有改变。此外,DR也显著地挽救了LT-HSCs比例的老化相关变化,但未能挽救LMPPs的减弱。

为了研究DR对HSC功能的影响,将上述处理小鼠中纯化的HSCs与竞争者细胞一起移植。有趣的是,DR衍生的HSCs导致全白细胞(WBC)和B淋巴细胞的嵌合度较高,B细胞的嵌合度/骨髓细胞的嵌合度之比也较高,骨髓细胞的嵌合度相似。

然而,临床HCT主要采用未纯化的造血细胞作为移植来源。虽然短期DR改善了HSCs本身的再生,但它也降低了HSCs的频率,这可能对HCT有负面影响。

值得注意的是,短期DR在骨髓移植(BMT)中表现出中性效应,表明短期DR实现的功能改善不能补偿HSCs频率的降低。

为此,研究人员进一步进行了另一个竞争性的BMT,使用更多的从DR供体取得的骨髓细胞,以达到与AL供体相同数量的HSCs移植。在这个实验中,DR供体的骨髓细胞在竞争性的连续移植中产生了更高的淋巴细胞和骨髓细胞嵌合体。

研究人员进一步研究了长期(9个月)DR对老年小鼠的影响。与短期DR相比,长期DR对HSCs和祖细胞的影响相似,CD150lowHSCs显著减少。

耐人寻味的是,长期DR增加了竞争性BMT中B细胞和骨髓细胞的嵌合体。此外,DR供体产生更高的B细胞/骨髓细胞中的嵌合体比例。值得注意的是,对淋巴细胞的有益作用早在BMT后1个月就开始了,这对移植后的早期抗感染能力至关重要。

从机制上看,长期DR使老化的HSCs中异常调控的线粒体通路恢复活力,并伴随着静止期HSCs的增加和DNA损伤信号的减少。短期DR对这些衰老标志的拯救也有类似的趋势,但程度要小得多。

综上所述,目前的研究表明,中期DR可改善老化HSCs的功能,长期DR甚至可改善骨髓移植中的造血重建,这在只有老年供者的情况下,可能对人类的HCT有相当大的意义。
