Atherosclerosis:动脉粥样硬化斑块惊现微生物

大量实验显示,在动脉粥样硬化斑块中监测到细菌DNA的存在,但细菌究竟可以稳定斑块还是促进斑块破裂目前仍不明确。本研究旨在研究粥样硬化斑块中的细菌DNA对动脉粥样硬化患者的发病有无影响,并研究无症状和症状明显的动脉粥样硬化患者的细菌种属差别。

大量实验显示,在动脉粥样硬化斑块中监测到细菌DNA的存在,但细菌究竟可以稳定斑块还是促进斑块破裂目前仍不明确。本研究旨在研究粥样硬化斑块中的细菌DNA对动脉粥样硬化患者的发病有无影响,并研究无症状和症状明显的动脉粥样硬化患者的细菌种属差别。

分别从无症状和症状明显的动脉粥样硬化患者上,通过动脉内膜切除术获取粥样硬化斑块模型,将标本分为3个不同部位分别对组织学易损性进行研究:A组取发生狭窄上游血管;B组取狭窄病灶本身;C组取狭窄发生下游血管。使用焦磷酸测序法对斑块内细菌DNA的16S rRNA基于进行检测,通过qPCR对细菌的总量进行测量。

通过对细菌16S rRNA进行qPCR分析,我们确认动脉粥样硬化斑块中的确存在细菌DNA,但是无症状和症状明显患者的3个检测部位之间细菌总量却无明显差别。未加权的UniFrac距离测量分析显示对不同症状的患者及取材的不同位置分析结果并无明显差别。对5种不同门类的细菌分别进行研究,发现占主导地位的两类细菌分别为变形菌门(48.3%)和放线菌门(40.2%)。在对不同细菌门类、种系、运算分类单位层面上的研究,发现不同症状患者、斑块不同部位之间无明显差异。

由此可得出,对无症状和症状明显的动脉粥样硬化患者及斑块不同部位的研究中,细菌DNA的数量及微生物构成比例对研究结果并无明显影响。这预示,对决定动脉粥样硬化斑块易损性研究上,还需从其他因素着手。
