结直肠癌(CRC)作为癌症相关死亡的第二大最常见原因,在全球范围内,每年约有1,800,000例新病例被确诊为CRC。尽管在CRC的早期检测和综合治疗方面已经取得了巨大的进展,但目前大多数晚期CRC患者的预后仍较差。

癌症的远端转移和复发仍是CRC患者死亡的主要原因。越来越多的新兴研究证实,多种基因和细胞通路参与了CRC的肿瘤发生发展和转移。因此,阐明潜在的分子机制或可为CRC患者提供有效的改善预后的治疗策略。


既往研究显示,CDX2参与了各种癌症的发生发展,并在癌症的侵袭和转移中起着至关重要的作用,但CDX2在CRC上皮-间质转化(EMT)进程中的作用目前还知之甚少。

该研究旨在通过免疫组织化学分析和一系列内外源实验,揭示CDX2在CRC侵袭和转移中的作用。


研究人员发现,CRC组织中CDX2的表达水平下调,而CDX2的减少对应着患者的不良预后。敲低CDX2能够促进结肠癌细胞的侵袭作用,并通过诱导EMT进程促进癌症的肝转移。

CDX2在CRC组织和正常组织中的表达

进一步的研究显示,CDX2通过抑制Akt和GSK-3β的磷酸化,降低Snail的表达、β-catenin的稳定和核转运。敲除β-catenin能够中和敲低CDX2对Slug和ZEB1的调节作用。机制研究显示,CDX2通过调节PTEN的表达以拮抗CRC中的PI3K/Akt活性,CDX2能够直接与PTEN的启动子结合并激活其表达。


综上,该研究首次发现CDX2可通过调节Snail的表达以及通过反式激活PTEN的表达来调节β-catenin的稳定性,最终抑制EMT进程和CRC的转移。
