Nature子刊:单细胞全基因组测序揭示人类心脏在衰老过程中的突变

该研究鉴定了人类心脏在衰老过程中积累的体细胞突变,研究结果或帮助我们理解心脏功能会如何随着我们年龄增长而退化。但仍需开展进一步研究验证作者鉴定出的过程是否和心脏衰老具有因果关系。

人在衰老过程中,组成人体(包括心脏)细胞的DNA会不断积累错误,这种错误也称体细胞突变。虽然有些体细胞突变似乎影响甚微,但另一些体细胞突变能诱发癌症,或有可能促进生理上的衰老。心肌细胞内积累的体细胞突变可能会促进细胞功能退化。不过,目前尚无法获得关于这些突变的数据。

2022年8月11日,波士顿儿童医院、哈佛医学院、Broad研究所等机构的研究人员在 Nature 子刊 Nature Aging 发表了题为:Somatic mutations in single human cardiomyocytes reveal age-associated DNA damage and widespread oxidative genotoxicity 的研究论文。

该研究使用单细胞全基因组测序技术鉴定了人类心脏在衰老过程中的体细胞突变,发现心肌细胞的体细胞单核苷酸突变(sSNV)会随着年龄的增长比许多分裂细胞和非分裂神经元以更快的速度积累。而且,心肌细胞具有提示DNA氧化损伤和DNA修复机制受损的突变以及碱基替换增加的突变特征,所有这些都会随年龄增长而累积

由于年龄积累的sSNV会产生许多破坏基因功能的突变,因此心肌细胞中的多倍体化可能提供一种遗传补偿机制,以最大限度地减少衰老过程中必需基因的功能缺失。与年龄相关的心脏基因突变积累为理解衰老对心脏功能障碍的影响提供了一个新范式。

在这项最新研究中,研究团队利用单细胞全基因组测序,分析了12名年龄在0.4岁到82岁个体的人心肌细胞的体细胞单核苷酸变异(sSNV),单核苷酸变异是指DNA序列中单个核苷酸发生突变。研究团队发现,心肌细胞具有提示DNA氧化损伤和DNA修复机制受损的突变以及碱基替换增加的突变特征,所有这些都会随年龄增长而累积
