腺苷(A)到肌苷(I)的RNA编辑给肿瘤转录组学中引入了许多核苷酸改变。但是由于转录后调节的复杂性,我们迄今为止仍然不知道RNA编辑对人类肿瘤中蛋白组学的多样性的影响。

为了解释这个问题,来自德州大学安德森癌症中心的科学家们在生物信息和计算生物学系副教授Han Liang、系统生物学系主任Gordon Mills教授的带领下对TCGA中的基因组学数据和CPTAC中的蛋白组学数据进行了综合分析。尽管收集数据的站点有限,但是研究人员还是发现A-I的RNA编辑可以通过改变氨基酸序列来改变了人类乳腺癌中的蛋白组学多样性。

研究人员在RNA和蛋白质水平确认了存在编辑,而编辑过的COPA蛋白质会增强癌细胞在体外的增殖、迁移和侵袭能力。

本文亮点:1)直接评估肿瘤组织中A-I的RNA编辑对蛋白组学多样性的影响;2)采用了一种更强大的计算机策略检测RNA编辑产生的各种多肽;3)独立的实验性证据表明RNA编辑诱导肿瘤中各种突变多肽;4)COPA编辑对癌细胞增殖、迁移和侵袭的影响。

总体来看,该研究表明了A-I的RNA编辑在肿瘤蛋白组学多样性中做出了重要贡献,同时强调了这一发现的临床转化价值。
