J Endod:不同牙本质处理剂对生长因子释放、间充质干细胞粘附和形态的影响

EDTA在牙髓再治疗中被认为是黄金标准。这篇研究的目的是为了评估不同牙本质处理剂(非EDTA)对生长因子释放、间充质干细胞粘附和形态的影响。

研究采用17% EDTA, 10%柠檬酸, 1%植酸(IP6)或37%磷酸处理牙本质片,最终冲洗后,通过脂肪来源的间充质干细胞(adMSC)定植3天。采用酶联免疫吸附测定定量分析牙本质片中所释放的TGFb-1, VEGF, BMP-2和FGF-2含量。从拔除的单根牙齿中制备40个牙根片段。采用SEM观察条件牙根片段上adMSCs的粘附和形态。通过1-way analysis对生长因子的定量数据进行统计学评估。

结果显示,柠檬酸处理后释放出最高比例的TGFb-1,其次为磷酸;2者无显著性差异,但是与EDTA和1% IP6相比,存在明显差异。酶联免疫吸附测定结果显示,在牙本质条件处理后,VEGF和FGF-2释放微弱,并且各组间无明显差异。1% IP6组中释放的BMP-2含量最高,但是各组间无明显差异。牙本质处理后adMSC定植培养3天显著增加所有生长因子的释放,并且与其他组相比较,磷酸是效果最佳的试剂。SEM结果显示,没有一种处理剂对干细胞增殖和在根部牙本质的粘附具有副作用。在牙本质条件处理组中,存在不同的细胞形态,如:圆形、椭圆形和扁平形,同时细胞粘附较好并且含有伪足。

结论:磷酸条件处理非常有用,并且可能对牙髓再治疗具有有益的作用。
