2019年底以来，2019冠状病毒病(COVID-19)在全球急速传播。核酸检测(NAT)在COVID-19患者诊断和治疗中发挥了重要作用。在某些情况下，56℃的热灭活策略被推荐用于在NAT之前对SARS-CoV-2灭活。然而，理论上，这种方法可能会破坏这种单链RNA病毒的核酸完整性，并在实时聚合酶链反应(RT-PCR)测试中造成假阴性。

本研究了热灭活是否会影响病毒NAT的结果。研究人员检测了热灭活对SARS-CoV-2定量RT-PCR结果的影响，特别是对低病毒载量标本的假阴性结果的影响。此外，还研究了不同标本类型、标本保存时间和化学灭活方法对NAT的影响。

研究显示，对确诊的COVID-19患者进行RT-PCR检测后，热孵育期Ct值升高。此外，在至少一次平行检测中，约一半的弱阳性样本(15个样本中的7个，46.7%)经热灭活后RT-PCR阴性。使用胍基裂解法保存标本对RT-PCR结果的影响较小，假阴性率更低(15个标本中有2个，占13.3%)，Ct值的升高明显小于热灭活法。

研究结果表明，热灭活对RT-PCR检测SARS-CoV-2的效率有不利影响。鉴于化学灭活化剂的适用范围有限，需要考虑其他确保对实验室人员进行全面保护的方法。
