NATURE:“万能”的CAR T还能逆转衰老

研究人员测试了以衰老细胞为靶点的嵌合抗原受体(CAR)T细胞作为有效的杀死衰老细胞手段的治疗理念。

细胞衰老的特点是稳定的细胞周期停止和分泌特征,以调节组织微环境。生理上,衰老作为一种肿瘤抑制机制,可以防止前恶性细胞的扩张,并在伤口愈合反应中存在有益作用。

从病理上讲,衰老细胞的异常积累产生炎症环境,导致慢性组织损伤,并有助于疾病发生,如肝脏和肺纤维化,动脉硬化,糖尿病和骨关节炎。因此,在小鼠中,从受损组织中消除衰老细胞可以帮助改善这些病理的症状,甚至促进长寿。

在这里,研究人员测试了以衰老细胞为靶点的嵌合抗原受体(CAR)T细胞作为有效的杀死衰老细胞手段的治疗理念。首先,研究人员确定尿激酶型质粒原激活物受体(uPAR)11是一种细胞表面蛋白,在衰老过程中被广泛诱导,并表明uPAR特异性CAR T细胞在体外和体内能够有效消融衰老细胞。

以uPAR为靶点的CAR T细胞可以延长接受衰老诱导联合药物治疗的肺腺癌小鼠的生存期,并恢复化学或饮食诱导肝纤维化的小鼠的组织平衡。

这些结果确立了靶向衰老细胞的CAR T细胞对衰老相关疾病的治疗潜力。
