Tissue Cell:铁皮石斛多糖诱导骨髓间充质干细胞分化神经元样细胞,改善大鼠脑卒中模型神经元功能

缺血性脑卒中目前临床治疗的方法是溶栓治疗和血管内介入,铁皮石斛多糖可促进骨髓间充质干细胞的神经元分化,并有效诱导神经元样细胞治疗缺血性脑卒中,这为缺血性脑卒中提供了新的治疗思路。

      脑卒中是一种常见的急性脑血管病,发病率、死亡率和致残率都很高。卒中主要由脑微循环异常引起,如脑内血管突然破裂或阻塞。溶栓治疗和血管内介入治疗是目前临床治疗缺血性脑卒中仅有的两种方法。然而,狭窄的治疗时间窗口限制了它们的使用。

      骨髓间充质干细胞来源于骨髓、羊水、胎盘、脐带等多种器官或组织,具有多种分化潜能和自我更新能力。动物实验的证据已证实,骨髓间充质干细胞移植可促进缺血性中风的功能恢复;也有研究报道了神经元样细胞移植治疗脑损伤。这为缺血性脑卒中提供了新的治疗思路。

     铁皮石斛是一种传统中草药,有证据表明铁皮石斛多糖(DOP)具有免疫调节、抗氧化、抗凋亡和抗肿瘤等生物活性和功能,但DOP对骨髓间充质干细胞向神经样细胞分化的影响尚未被研究。

     近期在Tissue&Cell发表的一项研究,基于以上发现,探索DOP是否促进骨髓间充质干细胞的神经元分化以及对缺血性脑卒中的作用。

 

      体外实验结果表明,DOP可促进骨髓间充质干细胞的神经元分化,其最佳浓度为25μg/mL,并且不会影响细胞增殖。Notch信号通路可能抑制、Wnt信号通路可能促进骨髓间充质干细胞的神经元分化。为了确定DOP诱导的神经样细胞是否具有神经功能,该研究建立了大鼠单侧瞬态MCAO模型,然后给予DOP诱导的神经样细胞。结果表明,DOP诱导的神经元样细胞移植可促进MCAO大鼠的恢复,并能有效诱导神经元样细胞治疗缺血性脑卒中

      然而该研究还存在一些局限,没有对注射的细胞进行标记和跟踪,没有检测到停止DOP治疗后诱导的神经元样细胞的表型是否稳定,这些细胞是否会整合到相关的神经回路中还缺乏直接证据。
