为探究驱动MCL病理发展的其他潜在机制,David Martín-Garcia等人采用荧光原位杂交(FISH)、全基因组/外显子组测序、基因表达和拷贝数序列等方法,对56例细胞周期蛋白D1-/SOX11+MCL展开研究。通过FISH在39(70%)例病例中发现CCND2重排,但未发现CCND3重排。研究人员通过全基因组/外显子测序对3例阴性的病例进行分析发现,在CCND3附近有IGK(2例)和IGL(1例)增强子劫持,与细胞周期蛋白D3过表达相关。

通过特异性的FISH探针,包括IGK增强子区域,研究人员在MCL中检测到10个额外的与CCND3(3例)和CCND2(4例)并列的隐藏IGK,这些病例分别过表达相应的细胞周期蛋白。

研究人员在4例无细胞周期蛋白D重排的细胞周期蛋白D1阴性的MCL病例组成的小子集中发现CCNE1和CCNE2上调。这些病例具有囊胚样形态、基因组复杂性高以及CDKN2A和RB1缺失。细胞周期蛋白D1阴性的MCL的基因组和基因表达谱均与细胞周期蛋白D1阳性的MCL无明显差别。

总而言之,所有细胞周期蛋白阴性的MCL均携带CCND2/CCND3与IG基因的重排,包括一种新的IGK/L增强子的劫持机制。具有侵袭性特征的细胞周期蛋白D1/D2/D3全阴性的MCL的亚群具有细胞周期蛋白E失调。特异性的FISH探针或可有助于对细胞周期蛋白D1阴性的MCL进行分子鉴别和诊断
