平滑肌细胞(SMC)在动脉粥样硬化中发挥至关重要的作用。芳烃受体(AHR)是一种环境敏感的转录因子,有助于血管发育,并与冠状动脉疾病风险有关。研究人员推测AHR可以通过调节SMC的表型调节来影响动脉粥样硬化,并开展实验对此进行验证。

研究人员采用单细胞RNA测序、染色质免疫沉淀等技术在动脉粥样硬化小鼠模型中研究AHR在血管疾病中的作用。

基因组研究与在培养的人冠状动脉SMC中的功能测定相结合后发现,AHR调节人冠状动脉SMC表型并抑制这些细胞骨化。在小鼠主动脉窦中的谱系追踪和活性追踪研究表明,Ahr通路在动脉粥样硬化病变斑块帽中的调节SMC中具有活性。此外,对SMC特异性Ahr敲除小鼠进行单细胞RNA测序显示,病变内膜的表达软骨细胞标记物(如Col2a1和Alpl)的调节SMCs的比例显著增加。我们称这些细胞为软骨细胞,也见于人冠状动脉的新生内膜中。

综上所述,研究人员推测,AHR可能具有保护作用:AHR促进了病变部位斑块帽完整性的维持,并减少了动脉粥样硬化组织中与疾病相关的SMC向软骨细胞的转化。。
