研究从未发育成熟的第三磨牙分离培养SCAPs。准备含不同浓度PRF (one eighth, one quarter, and one half PRF)的条件培养基。在施加PRF后,分别通过CCK-8和划痕实验检测SCAPs的增殖率以及迁移能力。茜素红染色检测矿化结节形成情况。WB检测SCAPs中成骨/成牙本质分化相关标志物以及ERK通路的表达情况。数据通过one-way analysis of variance进行统计学分析,显著性水平设置为P values <.05。

CCK-8结果显示,1/8的PRF能够显著促进SCAPs的增殖率和迁移能力(P < .005),而1/4以及1/2的PRF与对照组相比无明显差异。1/8和1/4的PRF能够促进SCAPs成骨/成牙本质分化标记物的表达以及矿化结节的形成。此外,PRF激活了ERK信号,ERK抑制剂明显减弱PRF诱导的SCAPs的成骨/成牙本质分化。

结论:PRF通过ERK信号促进SCAPs成骨/成牙本质分化;同时,PRF能够促进SCAPs的增殖和迁移能力,并且1/8浓度的PRF效果最佳。PRF对SCAPs良好的细胞生物学作用可能是PRF应用于牙髓再生疗法的基础。
