众所周知,怀孕期间,寨卡病毒(Zika)对孕妇的感染也会对胎儿造成破坏性的后果,包括了子宫内生长受限,小头畸形等。为了开发新的寨卡病毒感染的治疗手段,我们需要更好地了解寨卡病毒母婴传播的机制。 近日,华盛顿大学医学院的Indira U. Mysorekar课题组发文阐释了自噬途径在寨卡病毒垂直传播过程中起的重要作用。在人类滋养层细胞中,寨卡病毒的感染能够诱导细胞内的自噬活性。如果通过药物抑制细胞自噬,则能够抑制病毒的感染。此外,在体实验显示,在小鼠中,自噬基因Atg16l1的缺失会抑制寨卡病毒的垂直传播,减轻胎盘和胎儿的损伤,并在总体水平改善胎盘和胎儿的状态。这种保护作用是由于改变了胎盘滋养层细胞的自主自噬活性,而不是由于母体寨卡病毒感染情况的改变。现在有一种自噬抑制剂,羟氯喹,是在孕妇中批准使用的。该药物能够减轻寨卡病毒在胎盘和胎儿中的感染情况,减少胎盘和胎儿不良情况的发生。 因此,研究人员的工作解释了寨卡病毒垂直传播的新机制。基于自噬的治疗手段具有在人类中减少寨卡病毒母婴传播的潜质。 原始出处: Bin Cao et al.
