GUT:异常肠道菌群改变宿主代谢组并影响人类和啮齿动物的肾衰竭

方法:

研究人员将223名ESRD患者和69名健康对照人群的肠道微生物组,血清和粪便代谢组进行了相关研究。运用多维数据揭示这些数据集与慢性肾脏病(CKD)啮齿动物模型的使用之间的联系,以测试肠道微生物组对毒素积累和疾病严重性的影响。

 

 

结果

结果显示:一组富含ESRD的微生物与患者的临床变量紧密相关,并编码涉及毒素和继发性胆汁酸合成的功能;微生物功能的相对丰度与这些代谢产物的血清或粪便浓度相关。移植到肾损伤的无菌小鼠或经抗生素治疗的大鼠中的患者的微生物群比对照组的微生物群更能诱导更高的血清尿毒症素产生,加剧的肾纤维化和氧化应激。核梭杆菌能够提高尿毒症毒素的生产和促进肾脏疾病发展的CKD大鼠模型。益生菌动物双歧杆菌反而能降低这些物种的丰度,降低毒素水平和大鼠疾病的严重性。

 

 

结论

本项研究得出结论:ESRD患者肠道菌群异常,不利于临床代谢,提示肠道菌群将成为减少尿毒症患者的有希望的治疗目标。
