既往研究表明,与喂食低脂高纤维标准饮食(SD)的小鼠(100%)相比,高脂低纤维饮食(WD)喂养的小鼠经受手术损伤(切除30%肝)、抗生素和短期饥饿后的存活率降低(29%)。本研究旨在明确术前予以SD调理是否能逆WD喂养对肠道微生物菌群的影响并改善术后存活率

先采用WD喂养小鼠6周,并在暴露于抗生素、饥饿和手术前用SD进行饮食预康复(3天 vs 7天)。通过16S rRNA基因测序分析微生物菌群组成。用质谱法测定短链脂肪酸,采用16S基因扩增进行功能预测,以确定微生物菌群功能。

采用SD调理后24小时时,WD小鼠的饮食康复就恢复了粪便微生物菌群的细菌组成,由Firmicutes优势菌转变为拟杆菌属优势种。然而,在预康复期间(即3天),每种微生物量的粪便丁酸盐含量仍然很低,术后死亡率与纯WD喂养相比没有变化。微生物菌群功能分析显示,丁酸盐的减少可能是术后恢复失败的原因。相比之下,经过7天SD(7DP)调理后,丁酸盐产生菌恢复较好,小鼠的术后存活率也明显提高(WD vs 7DP vs SD:29% vs 79%vs 100%;p<0.001)。

总之,高脂低纤维饮食对肠道菌群的不良影响在改为低脂高纤维饮食后7天即可恢复。此外,粪便标志物或可成为提示肠道微生物菌群是否准备好承受手术等打击(包括接触抗生素和短期饥饿)的检测指标。
