Molecular Cell 杂志在线发表了中国科学院生物物理研究所张宏课题组题为The ER-localized transmembrane protein TMEM39A/SUSR2 regulates autophagy by controlling the trafficking of the PtdIns(4)P phosphatase SAC1 的研究论文,该文揭示了内质网定位的膜蛋白TMEM39A/SUSR2通过调节PtdIns(4)P的磷酸酶SAC1从内质网到高尔基体的运输,进而调控自噬活性的新机制。

细胞自噬(autophagy)是真核生物中高度保守的降解途径,指的是细胞通过形成双层膜的自噬小体,将其包裹的底物运送到溶酶体进行降解。多细胞生物自噬小体的形成和成熟过程需要多种蛋白的协同作用。张宏课题组前期以线虫为模式生物鉴定了一系列参与自噬小体形成和成熟过程的新基因,并将这些多细胞生物特有的自噬新基因命名为epg基因。张宏课题组近来利用线虫为模型,研究自噬活性在发育过程中是如何被调控的。这篇文章研究了通过遗传筛选鉴定的一个促进自噬活性的新基因,命名为susr-2。哺乳动物细胞中susr-2的同源基因编码内质网定位的跨膜蛋白TMEM39A。GWAS研究发现SUSR2的突变跟多种自身免疫性疾病的发生发展密切相关。但是TMEM39A的分子功能仍然完全未知。

这项最新的研究发现定位于内质网的跨膜蛋白TMEM39A/SUSR2通过调节PtdIns(4)P在细胞内的分布和水平来调节自噬活性。在SUSR2 敲减的细胞中,晚期内体/溶酶体上的PtdIns(4)P水平升高,促进了在自噬小体成熟过程中发挥作用的HOPS复合物的募集。进一步研究发现,SUSR2作为衔接蛋白与PtdIns(4)P的磷酸酶SAC1和COPII运输小泡的内层蛋白SEC23/SEC24相互作用,促进SAC1从内质网到高尔基体的转运。在SUSR2 敲减的细胞中,内质网上积累的SAC1水解PtdIns(4)P,伴随着PtdIns(3)P合成的增加,进而也促进自噬小体的形成。先前人们多关注PtdIns(3)P在自噬通路中的作用,该研究系统地阐明了PtdIns(4)P在自噬小体形成和成熟中的功能,扩充了人们对磷脂酰肌醇的全面了解,揭示了SUSR2在控制细胞内PtdIns(4)P的分布和水平参与调控自噬活性的重要作用。另外SUSR2还是多发性硬化(multiple sclerosis)和系统性红斑狼疮(systemic lupus erythematosus)等自身免疫性疾病的易感位点,这些研究为阐明这些疾病的发生提供了线索。

张宏为论文的通讯作者,张宏课题组助理研究员苗广艳和博士研究生张玉洁为论文的共同第一作者,张宏课题组博士研究生陈迪也参与了这项研究工作。该课题获得北京市生命科学前沿创新培育项目、国家自然科学基金、国家重点研发计划项目、中科院先导项目以及中科院前沿科学重点研究计划的资助。
